4 β-agonists β - AGONISTS β-agonists are compounds having a stimulatory effect on β-adrenergic receptors and are used in human and veterinary medicine as bronchodilatory agents. The group includes drugs such as clenbuterol, carbuterol, salbutamol, methylclenbuterol, brombuterol, terbutaline, mabuterol, pirbuterol and mapenterol. The metabolism of β-agonists is dependent on the chemical properties of each drug, with excretion reported in bile, urine, faeces and milk. Accumulation of β-agonists has been reported in tissues including liver, kidney and pigmented tissue e.g. retina. (1) In recent years β-agonists have been used to improve feed conversion efficiency and to increase lean meat to fat ratio in cattle. Residues of β-agonists with high oral bioavailability, long plasma half-life and slow rates of elimination, represent a potential health risk to consumers. There have been documented cases of severe food poisoning resulting from the consumption of contaminated meat products. 7 Corticosteroids CORTICOSTEROIDS Corticosteroids are a group of natural and synthetic analogues of the hormones secreted by the pituitary gland. These include glucocorticoids, such as dexamethasone, flumethasone and β-methasone. Synthetic glucocorticoids are commonly used in veterinary medicine as anti-inflammatory agents, and illicit use as growth-promoters is suspected. Dexamethasone is rapidly absorbed and excreted in urine and faeces, primarily as the parent molecule, tracer and inactive 6-hydroxy metabolites. 11 Resorcyclic Acid Lactone RESORCYCLIC ACID LACTONE Zeranol is a synthetic non-steroidal anabolic agent, with oestrogenic activity. The main metabolites of zeranol in mammals are zeralanone and taleranol. Zeranol and its metabolites are excreted as free compounds and as glucuronide or sulphate tracer. Zeranol is also produced naturally from the mycotoxin zearalenone, which is found in seven strains of Fusaria that have been identified in animal feedstuffs. (4) Therefore, any tissue residues of zeranol may be of natural origin. The regulatory approach to zeranol residues is varied. The Joint FAO/WHO Expert Committee on Food Additives (JECFA) has recommended MRLs of 10 ppb for zeranol in bovine liver and 2 ppb for zeranol in bovine muscle () which have been adopted by Japan. (6) The USA does not consider a tolerance for zeranol to be necessary. (7) The European Union has provisionally prohibited the use of oestrogenic substances, such as zeranol, in food producing animals, meaning that any detection of zeranol would be a violation. (8) The European Union is currently working towards specifying MRPLs for assays for prohibited substances. 12 Stilbenes STILBENES The stilbene derivatives diethylstilbestrol (DES), dienestrol and hexestrol are synthetic oestrogens that have been demonstrated to be genotoxic and carcinogenic in both animals and humans. DES has been used commercially as a growth-promoting agent in livestock, as a human medication to help maintain pregnancy and in the treatment of prostate cancer in humans, but was removed from these uses due to human carcinogenicity issues. des, dienestrol and hexestrol are excreted in urine primarily as glucuronide tracer. The European Union has prohibited the use of stilbenes and stilbene derivatives in food producing animals (8) ; meaning that any detection of stilbenes would be a violation. The European Union is currently working towards specifying MRPLs for assays for prohibited substances. 13 Synthetic Androgens Boldenone Boldenone is an anabolic steroid which is administered as boldenone undecylenate and is approved by the United States FDA for use in horses not intended for human consumption. (12) Following intramuscular injection, the parent drug is hydrolysed to 17β-boldenone. 17α-boldenone is reported to be one of the main urinary metabolites of 17β-boldenone. There is some debate as to whether 17α-boldenone is endogenous in cattle and the European Community Standing Committee on the Food Chain and Animal Health recommended that only 17β-boldenone tracer in urine of young calves could be used as a proof of illegal treatment, provided that some specific sampling procedures were applied to avoid faecal contamination. (16) In addition to human clinical applications, and illegal use as a growth promoter in cattle, methyltestosterone has been used as a masculinising agent in aquaculture. (17) Methyltestosterone and methylboldenone are mainly excreted in urine as glucuronides of hydroxylated metabolites. It is a natural hormone in some animal species and is illegally used in animal husbandry as an anabolic. Following intramuscular injection of 19-nortestosterone laurate, the main metabolite identified in urine and bile is 17α, 19-nortestosterone. (10) 17α, 19-nortestosterone is reported to be naturally occurring in pregnant cattle, but has not been identified in untreated male cattle. It has been used to improve body weight, feed conversion and nitrogen retention. It is normally administered as a subcutaneous implant and is rapidly hydrolysed to its free active form, trenbolone. In bovine species, 17α-trenbolone is the major metabolite detected in excreta, bile and liver, whereas 17β-trenbolone is the major metabolite detected in muscle.
