1 Validation and Development of Traps for Pests of Megachile rotundata Theresa Pitts-Singer 1 and Sam Malan 2 1 USDA ARS Pollinating Insects Research Unit, Logan, Utah 2 Biology Department, Utah State University, Logan, Utah A recent survey of Megachile rotundata cells from the western United States found an average of 24% of cells parasitized by wasps, and another 13% destroyed by predators (James and Pitts-Singer 2013). Wasps that invade nests while bees are nesting are cleptoparasites and parasitoids. Parasitoids may also infest cells during fall storage and summer incubation. Cleptoparasites include species of Sapyga and Chrysura, and parasitoids are species of Pteromalus, Tetrastichus, Monodontomerus, and Melittobia. There are also beetle predators of M. rotundata brood, including Trichodes, Nemognatha, and Tricrania that infest bee cells in the summer. (Other less common natural enemies of M. rotundata are not mentioned here.) To avoid the known substantial loss of brood due to natural enemies, methods of control should be employed, and some known techniques may need to be improved. Protocols to protect bees from some of these pests are applied during storage and incubation. The most common anti-parasitoid practices during storage and incubation are the well-timed use of dichlorvos strips and ultraviolet light (UV) with water/oil traps. However, UV light traps may not collect parasitoid wasps before they have laid eggs in new hosts. During the nesting season, parasitism can be limited by assuring a snug fit of backings on bee boards. Attempts to control cleptoparasites and predators that attack at the nest entrance has been the use of mechanical (unbaited) traps for Sapyga (Arnett 1980) or traps baited with the attractant cyclohexyl phenylacetate for Trichodes (Davis et al. 1979, 1983). These traps are not widely used (if at all) and largely assumed ineffective (personal communication with bee managers). Previous studies to prevent wasps and beetles from attacking M. rotundata were performed when wooden nesting boards were most common. Now many bee managers use polystyrene boards. Also, the assumed inefficiency of the known traps has diminished their use. In summer 2015, we examined the efficacy of Sapyga PVC traps and explored new avenues for controlling M. rotundata natural enemies. Objectives 1. Perform Y-tube bioassays to determine the attractiveness of a known attractant of Sapyga elegans (that attacks Osmia spp.) to S. pumila that attacks M. rotundata in North America. The chemical cue is decanoic acid. 2. Perform Y-tube bioassays to determine the attractiveness of odors from whole M. rotundata cell extracts to Sapyga pumila. 3. Perform field trials to test efficacy of two colors of PVC wasp traps, with and without whole cell extract added (i.e., a lure). 4. Perform field trials to test efficacy of two colors of polystyrene, screened parasitoid traps, with and without whole cell extract added (i.e., a lure). Methods: Laboratory Y-Tube Bioassays Olfactory Cues For the Y-tube bioassay, we used a simple apparatus (Fig. 1) (Pitts-Singer 2007) for performing multiple tests each day on three-day-old S. pumila females that had been maintained with males so that mating could occur. Experiments allowed wasps to choose between: 1) decanoic acid and a blank, and 2) whole cell extract and a blank. To make cues for testing, decanoic acid was purchased and dissolved in ethyl acetate for dispensing onto filter paper. Also, 100 M. rotundata whole cells (from which bees had emerged) were extracted with a methanol-chloroform solution which was applied to filter paper. Fresh chemical cues were made for each day of testing. Each wasp was observed in the Y-tube for up to 30 minutes to see which cue (i.e., arm of the Y) it followed. A choice was defined as the wasp actually touching, with antenna or legs, the muslin cloth that covered the end of the tube. Each female was tested only once. Significant differences between choices were determined using a Chi-Square analysis. 1

Results: Laboratory Y-Tube Bioassays Olfactory Cues For the decanoic acid vs blank test, 37 S. pumila females made a choice within the 30-minute test period, and 11 females did not choose. Their choices did not significantly differ between the decanoic acid and the blank (χ 2 = 1.32, d.f. = 1 p > 0.05). For the whole cell extract vs blank test, 21 S. pumila females made a choice within the 30-minute test period, and 24 did not. Choices made showed a significant preference for the whole cell extract over blank (χ 2 = 8.05, d.f. = 1 p < 0.005) (Fig. 2). Figure 2. Choices made by Sapyga pumila females in Y-tube tests. 2

3 Methods: Field Trials Trap Efficacy in the Field Standard PVC traps known to catch S. pumila were made from both black and white PVC pipes (Fig. 3). Prototype parasitoid sticky traps were made of small sections of polystyrene boards whose backing is a piece of sticky board. The front of the trap was covered with window screening so that only small insects could enter (i.e., no bees) (Fig. 4). Treatments for each trap type were color (PVC: black or white; sticky: black or grey) and whether they were baited with a lure made with the same whole cell extract as was used in the Y-tube bioassays. PVC traps with lures had pieces of impregnated filter paper dangled inside the pipe (Fig. 3). Sticky traps with lures had rolled, impregnated filter papers in 10 of the holes (Fig. 4). Figure 3. Four PVC traps (one each of both colors, with and without lures) were hung inside each of four M. rotundata shelters in two local commercial alfalfa seed fields in northern Utah. Four sticky traps (one each of both colors, with and without lures) were affixed to the ends of bee boards in the same shelters. Traps were collected and replaced with new ones each week from 10 July - 24 September, The identity and number of collected wasps were recorded. A generalized linear mixed model followed by a Tukey s multiple comparison test was used to compare effects of trap types, color, and if the trap contained a lure. Results: Field Trials Trap Efficacy in the Field Wasps collected in the PVC traps were Tetrastichus sp., Melittobia sp., and S. pumila. Of these wasps, the screening of the sticky traps only allowed Tetrastichus sp., and Melittobia sp. to be caught. Tetrastichus sp.: Enough Tetrastichus sp. were caught to compare between trap types in only Field 1. The PVC traps caught significantly more of these wasps than the sticky traps (F = 44.7; d.f. = 1, 3; P = 0.007) (Fig. 5). Within the PVC traps, there were significant effects of the lure and also the interaction of trap color and lure. Traps without the lure caught Figure 5. 4 significantly more wasps than traps with it (F = 7.2; d.f. = 1, 112; P = 0.009). Black traps without the lure caught significantly more wasps than both types of white traps and also the black traps with the lure (= 7.9; d.f. = 1, 112; P = 0.006). Color and lure had no effects on sticky trap efficiency in Field 1. Melittobia sp.: This wasp species was caught in enough traps in both fields to look for trap type effects. The PVC traps caught significantly more Melittobia sp. than the sticky traps (F = 240.8; d.f. = 1, 3; p = ) in both fields (Figs. 6&7). For Field 1, sticky traps without the lure present caught significantly more Melittobia sp. than traps with the lure (F = 5.6; d.f. = 1, 126; P = 0.020), but color had no effect (Fig. 6). In Field 2 sticky traps, the color lure interaction was significant (F = 4.1; d.f. = 1, 132; P = 0.05), but the Tukey s test showed no significant comparison, and the standard errors were very large for this data set. 5 Sapyga pumila: These wasps were only trapped in the PVC traps of Field 2. Significantly more female and male wasps were caught in the black traps compared to the white ones (females: F = 18.2; d.f. = 1, 126; P < , males: F = 6.9; d.f. = 1, 126; P =0.01) (Figs. 8&9). Presence of lures had no significant effect. Discussion: New findings from this first year of study offer important insights into the utility of traps used to control pests of M. rotundata. In situations where a comparison between PVC and sticky traps was possible, we found that the PVC traps were more efficient at catching parasitoid wasps than the sticky traps. We do not understand why this style of trap caught more wasp species than Sapyga, for which it is designed. Perhaps the orientation and location of the traps within the shelter and their shapes likely are factors that we should consider or manipulate in future studies. Decanoic acid was previously discovered to be attractive to nest-seeking Osmia lignaria females, which are also megachilid bees. Furthermore, this compound is attractive to its cleptoparasite S. elegans, possibly indicating a host location cue. We found that the M. rotundata cleptoparasite S. pumila females were not attracted to decanoic acid. Thus, we explored the use of cues from the host of S. pumila, which might be found in M. rotundata cells from which adults had emerged. Simple application of whole cell odor cues (in the one amount that we chose) to filter papers inserted into PVC and sticky traps did not result in a higher catch of any wasp species compared to those traps without the lures. In fact, the catch sometimes was reduced in traps that contained whole cell odors. Perhaps these pest wasps are attracted to the cell odors only for long-range orientation, but cues for finding viable host cells require other visual or tactile cues, such as cocoon or larval odors, pollen-nectar provisions, or freshly cut alfalfa leaves. Color attraction differed between wasp species. More Tetrastichus were caught in black PVC traps, while Melittobia were found more frequently in the white traps. More S. pumila were caught in the black traps, as was expected according to knowledgeable bee managers (personal communication). We plan to continue this line of research by: 1) exploring other chemical cues as lures and testing more wasps species, and 2) changing the design and location of traps. We also will test the efficacy of the different trap types during management of the bees before and after nesting (i.e., incubation and storage). It will be very interesting if we find that different pests require different trap types and cues for the best control strategies. 5
