IonSense is not responsible for the misuse of this instrument or the ignoring of the safety guidelines outlined in this manual. Safety Labels: Risk of Electrical Shock; ID-Cube Source The ID-Cube Source contains high voltage lines that carry as much as - 6000V and pose an electrical shock hazard to untrained persons. Disconnect AC power and HV cabling before opening and servicing source. 1

Risk of Electrical Shock; ID-Cube Power Supply There are no user serviceable parts within the ID-Cube Power Supply and users should not remove the cover. The power supply contains electrical lines that carry as much as 5A and pose an electrical shock hazard to untrained persons. Open Spot Card Mesh Screen: Risk of Electrical Shock and Hot Surface: ID-Cube Source When the OpenSpot Cards are inside the source and in use the mesh screen could have a maximum of 5A of current applied to them, and could reach temperatures beyond 500C. 2

The ID-Cube should never be operated without the MS Interface being attached. Without the interface (pictured on the left) hazardous electrical current and hot surfaces are accessible. The interface blocks this access (pictured on right). No tools or other items should be placed inside the Open Spot card slot during ID-Cube use due to risk of shock or heat damage related to the mesh screen. 3

Proper Use and Handling of Gas and Gas Cylinders: Users should know and understand standard gas cylinder handling safety guidelines. See the gas section of this manual for instructions on the proper installation of gas for the ID-Cube. Certifications: RoHS: The ID-Cube is RoHS compliant. CE: The ID-Cube has been CE Certified for European Standards. Our DoC (Declaration of Conformity) can be found on our website at the following link:: http://www.ionsense.com/regulatory The DoC lists the Safety and Electrical Directives, and the tests within those directives that the ID-Cube was subject to. Electrical Safety Installation Category: The ID-Cube is a Category I installation, which is defined as: Equipment having a Ground Pin connection and receives its AC power from a protected (circuit breaker or fused) source having isolation. 4

Operating Environment: Temperature Range The laboratory room temperature must be maintained between 50 and 90 F (10 and 32.2 C). The laboratory must have good air flow in the vicinity of the ID-Cube and must maintain a constant temperature within the ranges stated above. Humidity Relative humidity in the laboratory room must be in between 30 and 70%. Vibrations Floors must be free of vibrations caused by other equipment in nearby locations. Power The ID-Cube is designed to accept 100-240V~ (1.1A) at 50/60 Hz (55W). Space requirements ID-Cube Vapur Flange: The Vapur attaches directly to the MS Interface. The flange will occupy the entire interface area and extend out a depth of 3-4. ID-Cube Source: The source attaches to the Vapur flange. Its maximum dimensions are: (w x h x d) 4 x 8 x 8 ID-Cube Power Supply: The power supply attaches to the source via cable; whose length is approximately 3 feet. The power supply must sit on its rubber feet. Its foot print is 6.5 x 5.5 with a height of 2. 5

Particulate Matter: Air in the laboratory room must not have an excess of dust, smoke, or other particulate matter. Gas Requirements The ID-Cube can use either Nitrogen or Helium gas, depending on the desired analysis. The recommended grades are: Helium: at least 4.7 grade (99.997% pure.) Nitrogen: at least 4.8 grade (99.998% pure.) Recommended size of tank is 300L. 6

Mounting ID-Cube Source: Installing the Flange: On all mass spectrometer instruments, an adapter flange is required to mount the ID-Cube. Please refer to the manual for the manufacturer specific flange required. ID-Cube Source: Once the flange is properly installed, the ID-Cube Source can be attached to the interface. The base plate will attach directly to the underside of the flange. Place the two provided M6 screws through the holes in the bottom of the base plate and screw into the flange using a 5 mm Allen wrench. 7

Connecting the ID-Cube: NOTE: There is a quick reference guide of ID-Cube connections features, and settings at the end of this document. It may be useful to reference that index during installation. High Voltage: High voltage (HV) for the ID-Cube Source is supplied from the mass spectrometer. The cable may be connected directly to the MS or to the Vapur Interface; reference the flange specific instructions for more details. Once the cable is connected on the MS side, plug the opposing end into the ID-Cube connector labeled Mass-Spec High Voltage. 8

The HV cable has a locking nut that can be screwed forward. When in the forward position the cable cannot be removed from the bulkhead. Be sure to screw it back before attempting to remove cable from source. Open position is show on top, and locked on the bottom. 9

Gas: The ID-Cube has one gas input for either Nitrogen or Helium gas. The ideal gas depends on your experiment and the desired conditions. The gas input is 5/32 Press-to-Fit for PTFE tubing. The standard tubing used is PTFE 4mm OD with 1mm ID; 25ft is included with each ID- Cube. Below, the gas input is shown: Cut an appropriate length of PTFE tubing to reach from the ID-Cube Source to the gas supply. Be sure to use the provided tube cutter; this is important as a clean straight cut will ensure minimal leakage from the press-to-fit connection. 10

Attach one end to the gas regulator using the fittings provided. If the regulator has a female end, use the ¼ hex nipple to convert to it to male. Attach the other end of the tubing to the ID-Cube source. Be sure the tubing is pushed all the way in. NOTE: The regulator pressure for both Helium and Nitrogen should be set to 75-80 PSI (5.5 bar or 0.55MPa). This ensures optimal gas flow and performance. 11

Power Supply Output: The ID-Cube Power Supply provides the power needed to heat the mesh screen in the Open Spot Cards. Plug the Power Supply Output cable into both the ID-Cube source and the power supply. NOTE: The end of the cable with the ferrite attached (black rectangular clamp) should plug into the Power Supply. The other end goes into the source. AC Power: Next, connect the AC power cable for the power supply into a standard wall socket. The power supply accepts 100-240VAC (1.1A) and 50/60HZ (55W). 12

Operating the ID-Cube: There are 4 overall steps in the operation of the ID-Cube. The order of these steps matters and should be done as shown below. Vapur Pump: The ID-Cube Vapur Flange requires a pump to operate. This pump will remove excess Helium and Nitrogen from the entering the MS. Connect the IonSense provided valve to the pump, and the rubber tubing to the Vapur Flange. Once this is complete turn on the pump; there should be a noticeable suction on the ceramic tube inlet attached to the Vapur flange. NOTE: Turning on ID-Cube gas flow before powering on the pump may cause the MS to lose vacuum as it will not be able to pump out the amount of Helium the ID-Cube is flowing. 13

Gas Flow: Gas Flow Rate can be controlled on the back of the ID-Cube using an analog flow controller. Turn the knob to the right to increase gas flow to the desired amount. The ball inside will signify the flow rate on the printed scale. The units printed directly on the scale are SCFH (Standard Cubic Feet per Hour) and the device is calibrated for Air/Nitrogen use. However, these units can be converted to L/min for either Nitrogen or Helium. Refer to the Gas Conversion Chart in the Reference Guide of this manual for the flow rate values. 14

High Voltage: Now that gas is flowing, the HV can be turned on. The high voltage used for the ID-Cube is turned on and off via the MS software. Refer to the MS specific manual for more details. NOTE: Turning on HV without gas flow will cause damage and corrosion to the internal components of the ID-Cube source, and will reduce sensitivity and life of the product in the long term. Sampling: Open Spot Cards: Once gas is flowing and the HV is on the Open Spot Card can be prepared for use. NOTE: Do not touch the metal screen in the Open Spot Cards with bare fingers, as this will cause contamination of the sampling area and can affect results. First spot the card with the desired analyte, then squeeze together the handles next to the slot to open up the internal card clamp. 15

Place the Card into the slot and release the handles. Power Supply: With the card in place, the power supply can now be used to heat the screen and complete the analysis. The power supply has 3 settings (low, med, high) which correlate to the maximum temperature the screen will reach. 16

The power supply output works as follows once the Start button is pushed: A 1 second wait at 0 Amps for the contact closure Then a 20 second linear ramp from 0 to the selected setting Then a 10 second wait period at the selected setting. The appropriate settings are the following: Low: 3 Amps - this setting permits analysis of high- and mediumvolatility components such as flavor volatiles. The results from using the "Low" setting are similar to those observed when operating the DART-SVP with its gas temperature set to 200 degrees Centigrade Medium:4 Amps - this setting permits analysis of medium-volatility components such as small molecule pharmaceuticals, fatty acids, and sterols. The results from using the "Medium" setting are similar to those observed when operating the DART-SVP with its gas temperature set to 350 degrees Centigrade High: 5 Amps - - this setting permits analysis of low-volatility components such as triglycerides, trimethylsilyl-derivatives of normally non-volatile materials such as carbohydrates, and commercial dyes for food and cosmetics. The results from using the "High" setting are similar to those observed when operating the DART-SVP with its gas temperature set to 450 degrees Centigrade NOTE: The temperature of the metal mesh of the OpenSpot card is influenced by the flow of ionizing gas through its porous structure. Thus the actual temperature of the desorption will be influenced by the flow rate of the gas and the vacuum pull of the VAPUR interface. 17

Select the appropriate setting and push the button on the supply. While the green indicator light is active current is still moving through the screen. Once the light turns off the heating is complete, and the card can be removed. Shut Down: The shutdown sequence of the ID-Cube should be the reverse of the setup sequence. Sampling: Remove any sampling cards still in the Open Spot Card slot. High Voltage: Turn off the High Voltage to the ID-Cube source using the MS software. Gas Flow: Reduce the gas flow to zero on the ID-Cube source using knob on the back. Vapur Pump: Turn off the pump to the Vapur Flange. 18

Trouble Shooting and Maintenance: Fuses: There are two fuses inside the ID-Cube Power Supply. Fuse Specifications: 2A, Slow Blow, PN: 2300002.HXP NOTE: These fuses are NOT user or field replaceable. If they are believed to be blown then please contact IonSense Service. Spillage: If any hazardous materials are spilled on the ID-Cube during operation then contact IonSense regarding necessary decontamination instructions and materials. Do not attempt to clean ID-Cube without contacting IonSense to approve of decontamination agents that will not damage or affect the performance and reliability of the ID-Cube. Ceramic Tube: The ceramic tube used in the Vapur flange can be cleaned in an oven or furnace if it was contaminated by samples or a spillage. IonSense recommends a temperature no higher than 750 C to ensure the ceramic will not crack. Service Contact Information: IonSense Inc. T: 781-231-1739 Email: service@ionsense.com 19
