Growth Promoter Arrays Growth-promoting drugs have been used by the livestock industry for over 30 years to improve an animal s ability to efficiently use nutrients and produce leaner, more affordable meat. Concerns over the use of hormones in food-producing animals first arose when a number of incidents linked hormone residues in meat with various medical conditions after consumption. As a result, the use of growth-promoting hormones in livestock production is either prohibited or strongly regulated. Compliance with these regulations is ensured by national monitoring programmes. Sample preparation Multi-analyte sample preparations for urine, tissue and feed are provided for use with this unique multiplex system. Due to the nature of this collection of growth promoter compounds, multiple sample preparations would be required. Randox however offer one single preparation to cover all 9 of these compounds (35 including cross reacting analytes). The methods provide a simple clean up without the use of costly solvents. URINE PREPARATION Dilute centrifuged urine 10 fold in assay diluent and apply directly to the Biochip MULTIPLE MATRIX SCREEN From 5ml of a centrifuged and filtered urine Deconjugation and neutralisation Dilution and centrifugation TISSUE PREPARATION From 1g of homogenised sample Addition of neutral aluminium oxide along with methanol/1m NaOH Sample is vortexed, rolled and centrifuged Supernatant is decanted Process repeated with remaining pellet Supernatants are frozen, centrifuged and diluted with column wash buffer FEED PREPARATION From1g ground feed sample Addition of methanol/h2o Sample is vortexed and centrifuged Supernatant is decanted Addition of extraction buffer vortex and centrifugation Supernatant is decanted 125µl of each supernatant are then combined and diluted in wash buffer SAMPLE IS NOW READY FOR APPLICATION TO THE IMMUNOAFFINITY COLUMN SAMPLE PURIFICATION - IMMUNOAFFINITY COLUMNS (IAC) Sample clean up is an essential part of drug residue testing, whether it is performed by ELISA, LC/MS or any other technique.
