The transparent grip head allows the user to visually monitor the position of the capillary. The capillary is fixed in the universal capillary holder via the grip head. 8 CellTram Air CellTram Oil CellTram vario 2 Product description 2.2 Features The CellTram Air, CellTram Oil and CellTram vario are manual piston pumps. Using a movable piston, the CellTram generates differences in pressure which are transferred directly to a capillary via a pressure tube. The piston is moved in the CellTram using the dial. A clockwise rotation, i.e. to the right, results in an increase in pressure, the capillary contents are pressed out. A counterclockwise rotation, i.e. to the left, results in a decrease in pressure, aspiration into the capillary takes place. To assist fine pressure adjustment, the dial has a scale. The capillary is inserted into a capillary holder which in turn provides the connection to the pressure tube. The universal capillary holder enables permanent mounting of various capillaries in the laboratory. The CellTram Air, CellTram Oil and CellTram vario have a very ergonomic design which enables easy handling, particularly in conjunction with all control units of Eppendorf micromanipulators CellTram Air The CellTram Air uses air for pressure transmission. This guarantees clean and simple handling of the piston pump. A complete revolution of the dial corresponds to a change in volume of approx. 88 μl. The position of the piston is displayed in the piston window on a scale from 0 to 15 mm corresponding to the strokes CellTram Oil / CellTram vario The CellTram Oil and CellTram vario use oil for pressure transmission which, unlike air, can hardly be compressed. This ensures its operating cycles are very controlled and constant. The simple filling technique reduces the filling time and simplifies oil filling. To ensure controlled, constant pressure transmission, the cylinder, pressure tube and part of the capillary are filled with oil. A complete revolution of the dial (coarse drive) corresponds to a change in volume of approx. 9.6 μl. The CellTram vario has an additional fine drive dial. The transmission ratio between fine drive and coarse drive is 10:1. A revolution of the fine drive corresponds to a change in volume of approx μl. This enables ultra-fine cell movements. The position of the piston can be observed in the transparent cylinder. The filling bore with the plug screw is at the top of the cylinder. The pressure tube is connected to the CellTram Oil/ CellTram vario with the aid of the tube connection screw. The pressure tube has a joint at both ends and thus connects the CellTram Oil/CellTram vario and the universal capillary holder. 8

Before using the device, read the operating manual carefully and familiarize yourself with the device's mode of operation. 3.2 Intended use The CellTram has been designed and manufactured for use in the field of Intracytoplasmic sperm injection (ICSI) in conjunction with Eppendorf VacuTips and Transfertips. The CellTram is thus considered a medical device within the scope of Directive 93/42/EEC. The CellTram is intended exclusively for indoor use, and for operation by qualified staff. 3.3 Warnings for intended use WARNING! WARNING! Risk of injury from flying glass capillaries and splinters. At high pressure, capillaries may become detached from the grip heads and be ejected at high speed, or they may shatter if not handled properly or they come into contact with the petri dish or other objects. Always mount the capillaries without pressure. Never point capillaries at persons. Always wear protective goggles. Make sure that the capillary does not come into contact with the petri dish or other objects. Check that the capillary is fixed correctly in the grip heads. Check that the outer diameter of the capillary corresponds to the specifications of the grip head. Check that the capillary is not blocked. In the case of independently-manufactured capillaries: make sure that the capillary is capable of withstanding the applied pressure. CAUTION! CAUTION! Poor safety due to incorrect accessories and spare parts. The use of accessories and spare parts other than those recommended by Eppendorf may impair the safety, function and precision of the device. Eppendorf cannot be held liable or accept any liability for damage resulting from the use of incorrect or non-recommended accessories and spare parts or from the improper use of such equipment. Grip head 0 is for capillaries with an outer diameter of 1.0 mm to 1.1 mm only. Other capillaries may only be used with the corresponding grip head (see Ordering information) (see Accessories on p. 25). 1. Screw the pressure tube on the CellTram. 2. Screw the pressure tube on the universal capillary holder. 3. Mount the capillary (see Mounting the capillary on p. 17). 4.2 CellTram Oil / CellTram vario - filling oil Hint! Eppendorf recommends that you work with an air cushion in the visible area of the capillary in order to avoid contact between the oil and the medium. Hint! Observe the legal regulations for purity of the oil. The supplied oil is generally used in the ICSI area to coat drops containing embryos / egg cells or sperm. The supplied oil is not sterile. Additional instructions can be found in the safety data sheet of the manufacturer. Hint! Aspirating oil with the syringe may lead to the formation of tiny air bubbles. This should be avoided as these air bubbles may be transferred to the CellTram which may impair or slow down the filling process. If the syringe contains air bubbles, these should be collected by long-term storage. Large air bubbles can be pressed out during filling. 11

Turn the dial to move the piston to the initial position. 2. Screw the pressure tube on the CellTram. 3. Screw the pressure tube on the universal capillary holder. 4. Screw the filling tube to the syringe using the Luer Lock connection. 5. Aspirate at least 3 ml of oil into the syringe. 6. Unscrew the filling screw from the filling bore by hand. 7. Screw on the filling tube to the filling bore. 12

During the first filling, several air bubbles will probably be present between the piston and the cylinder. 10. Allow these bubbles to rise by moving the dial or the coarse drive forwards and backwards. 13

14 CellTram Air CellTram Oil CellTram vario 4 Installation 11. Using the syringe, fill the oil completely into the CellTram. Make sure that there are no air bubbles in the tip of the piston chamber. 14

Visually check that no air bubbles remain in the universal capillary holder. 14. Return the CellTram to a horizontal position. 15. Unscrew the filling tube. 15

16 CellTram Air CellTram Oil CellTram vario 4 Installation 16. Using the syringe, fill the filling bore with oil. 17. Screw the filling screw by hand into the filled filling bore. When doing so, make sure that no new air bubbles enter the bore. 18. Soak up any excess oil using a soft cellulose cloth. Hint! Make sure that the piston is positioned behind the filling bore when filling and refilling with oil. 19. If the CellTram still contains air bubbles: hold the CellTram at an angle to move the air bubbles to the filling bore. Remove the air bubbles. 20. Mount the capillary (see Mounting the capillary on p. 17). 16

Make sure that the capillary does not come into contact with the petri dish or other objects. Check that the capillary is fixed correctly in the grip heads. Check that the outer diameter of the capillary corresponds to the specifications of the grip head. Check that the capillary is not blocked. In the case of independently-manufactured capillaries: make sure that the capillary is capable of withstanding the applied pressure. CAUTION! CAUTION! Risk of injury when unpacking the capillary If the packaging of the capillary is removed incorrectly, it can lead to cuts and damage to the capillary. Do not reach into the protective tube of the capillary. Hint! Grip head 0 is for capillaries with an outer diameter of 1.0 mm and 1.1 mm only. Other capillaries may only be used with the corresponding grip head (see Ordering information) (see Accessories on p. 25). 1. Slightly screw the grip head into the universal capillary holder. 2. Slide the capillary through the inner O-rings up to the stop into the universal capillary holder. Hint! There will not be a perceptible stop, when using microcapillaries with an inner diameter of <0.8 mm. 3. Tighten the grip head. 4. In the case of CellTram Oil and CellTram vario, carefully wipe off any excess oil from the universal capillary holder and the capillary with a soft cellulose cloth if necessary. When using Eppendorf Femtotips 1. Screw the Femtotip adapter into the universal capillary holder. 2. Screw the Femtotip into the Femtotip adapter. 4.4 Setting the angle of the CellTram 1. Loosen the fastening screw in the tilting joint with an Allen key (4 mm). 2. Set the CellTram via the tilting joint. 3. Tighten the fastening screw in the tilting joint with an Allen key (4 mm). 17

CellTram Air: An overpressure or vacuum can be built up in the capillary using the volume of the piston chamber (cylinder) and the pressure tube. Therefore, the pressure behavior varies depending on the position of the piston. If the piston is located, e.g., in position 10 to 15, the differences in pressure are greater than, e.g., in position 0 to 5. By carefully selecting the position of the piston, the sensitivity of the CellTram Air piston pump can be adapted to your requirements. 1. Fix the universal capillary holder into the tool holder of the Eppendorf micromanipulator. 2. Determine the correct length of the capillary by moving the universal capillary holder in the tool holder. 3. Determine the correct rotation of the capillary by turning the front knurled screw. 4. Move the micromanipulator upwards (see Operating Manual of the micromanipulator). 5. Position the petri dish. 6. Move the capillary into the medium and ensure its correct positioning by observing it under a microscope. Establishing pressure equilibrium in the capillary 7. You have three options: Wait until equilibrium has been established. Turn the piston clockwise (i.e. to the right) to establish a counter pressure which compensates the active capillary force. Only CellTram Air: loosen the ventilation screw. Tighten the screw again after pressure compensation. 8. Move the capillary near to the desired cell. 9. Generate a vacuum by turning the dial counterclockwise (to the left). Hint! In hold mode: Excessive constant pressure can damage the cell. After the cell has reached the capillary tip, an optimal constant pressure should be set. Hint! In transfer mode: Ensure that pressure is equilibrated sensitively and evenly during cell aspiration into the capillary. 10. While the cell moves towards the capillary: it may be necessary to turn the dial clockwise (to the right) to slightly equilibrate the vacuum generated in order that the cell is not fixed too firmly to the capillary tip. 18

19 CellTram Air CellTram Oil CellTram vario 5 Operation 5.2 Only CellTram Air: Aspirating liquids 1. Dip the capillary into the liquid that is to be aspirated. 2. Turn the dial counterclockwise to the left (by approx. 2 revolutions) to aspirate the liquid by creating a low pressure. 3. Wait for a short period and then loosen the ventilation screw. 4. Retighten the ventilation screw after pressure compensation. 19

Remove the O-rings and the sealing washer with the extraction tool. 2. Insert the 2 new O-rings and a new sealing washer in the correct order. 3. Make sure that the 2 sealing washers and 2 matching O-rings are correctly arranged in the grip head. 6.2 Cleaning NOTICE! NOTICE! Damage from the use of aggressive chemicals. Do not use any aggressive chemicals on the device or its accessories, such as strong and weak bases, strong acids, acetone, formaldehyde, halogenated hydrocarbons or phenol. If the device becomes contaminated with aggressive chemicals, clean it immediately with a mild cleaning agent. Hint! Choose the disinfection method which corresponds to the legal determinations and guidelines in place for your range of application. If you have any further questions regarding the cleaning and disinfection or decontamination, or regarding the cleaning fluid to be used, contact the Eppendorf*AG Application*Support. The contact details are provided on the back of this operating manual. 1. Clean the CellTram, pressure tube and universal capillary holder with water or a mild lab detergent. 2. To remove residual liquid, blow out the pressure tube and universal capillary holder with compressed air after cleaning. Hint! There must not be any residual liquid remaining in the pressure tube or in the universal capillary holder after cleaning as otherwise steady pressure behavior will be compromised. 20

Do not use UV, beta, or gamma radiation or any other high-energy radiation source for disinfecting. NOTICE! Damage to the device due to autoclaving. Do not autoclave the CellTram. Required equipment Alcohol (ethanol, isopropanol) or disinfectants containing alcohol Proceed as follows: 1. Select a disinfection method that complies with the legal regulations and guidelines applicable to your range of application. 2. Wipe all surfaces of the CellTram and the equipment with the disinfectant. 3. Flush the pressure tube and universal capillary holder with the disinfectant if required. 4. To remove residual liquid, blow out the pressure tube and universal capillary holder with compressed air after cleaning. Hint!
