1 Preface DGT-CO2BX is a Digital Control Unit that generates a CO 2 -Air mixture with an adjustable CO 2 concentration in the range 0-20%, with an accuracy of ± 5% of CO 2 concentration, for instance, if CO 2 set point is 5%, accuracy is ± 0.25%. DGT-CO2BX can be used as a stand-alone device as well as in combination with any kind of microscope incubator. The air flow is regulated by a floating ball air flow meter in the range 0.2 0.8 Nliters/min. A CO 2 infra red sensor continuously measures CO 2 concentration in the mixed gas stream and a PID closed loop controller gives feedback to a fine valve regulating CO 2 flow. Measured value of CO 2 concentration is displayed in real time in green (DGT-CO2BX- PLUS displays also set point value). An alarm can be set on CO 2 concentration, so that, if CO 2 concentration goes below such value, display colour turns red and optionally a buzzer turns on. In the model DGT-CO2BX-PLUS the serial RS-232 interface and the CO 2 Control Software allow the user to control the unit with a personal computer and to acquire and store concentration data in computer memory. The DGT-CO2BX, CO 2 Digital Controller, together with the Okolab Microscope Stage Incubation Systems is designed to maintain all the required environmental conditions for cell cultures right on the microscope stage, thus allowing prolonged observations of cell events. 2

4 Equipment 4.2 Equipment supplied The DGT-CO2BX (-PLUS), CO 2 Digital Controller, is comprised of the following components: Hardware: #1 device, DGT-CO2BX (-PLUS-S) Software (Optional): Plastic connection tubings kit + accessories Electric Cables 4.2.1 Hardware details #1 CD-ROM with DGT-CO2BX-PLUS-S Software 1) DGT-CO2BX Version, Digital CO 2 Controller. CO 2 can be regulated in the range 0-20% with great accuracy. Measured CO 2 value is displayed in real time. 2) DGT-CO2BX-PLUS Version, Digital CO 2 Controller Plus. Same as DGT-CO2BX + RS232 serial port to control and read CO 2 levels via software. 4.3 Equipment NOT supplied Personal Computer (OS: Windows 2000 /Xp) with #2 RS232 Serial Communication Ports or #1 USB Port. CO 2 reservoir. Air reservoir (It can be substituted by a compressed air line or External Air Pump) Pressure Gauge for Air with regulator, degree of filtration 20μm and condensate drain (provided by Okolab on request). 6

4.4 Digital CO 2 Controller setting Depending on you chose of the Okolab Digital CO 2 Controller version, the Front and Rear will be different. Tip DGT-CO2BX and DGT-CO2BX-PLUS versions only differ on the Display Screen and on the presence of the RS232 serial communication port on the Rear. 5 Connections 5.2 Set the gas lines Provide your system of CO 2 reservoir with safety valve and pressure gauge with pressure rigid tubes 6 mm OD exiting (Figure 3.number 1). Provide your system of Air reservoir (Okolab suggests to use a compressed Air line) with safety valve and pressure fine gauge with pressure rigid tubes 6 mm OD exiting (Figure 3.number 4). If a compressed air line is not present in the lab an Air Pump with filter inside can be used. It is not included in the system, upon request (Figure 3.number 5). Provide your system of O 2 instead of Air (optional configuration) reservoir, or compressed line, with safety valve and pressure gauge with pressure rigid tubes 6 mm OD exiting (Figure 3.number 4). Mount the Pressure Gauge for CO 2 with regulator, degree of filtration 20μm and condensate drain (supplied) on the wall vertically-(figure 3.number 2). Be sure that Air stream is FILTRATED and with a STABLE Pressure Gauge otherwise mount another Pressure Gauge for air with regulator, degree of filtration 20μm and condensate drain (non supplied, if you need ask to Okolab for a prompt shipping)- (Figure 3.number 6). If there is no pressurised air line in the lab an Air Pump with filter inside can be used. Follow the arrow on the Pressure Gauge for the correct gas In-Out. Blue rigid tube with 6mm O.D., 3m long Figure 3: Gas lines scheme from reservoirs to Control Unit 5.3 Tubes connection 5.3.1 Connect DGT-CO2BX with Humidifying Module or Specimen Chamber Connect the Blue marked tubing #1 tube composed by 1 meter of blue rigid tube of 6mm OD at each end, 50cm of silicon tube of 4mm I.D. with a filter, degree of filtration 0.2 micron. listed before to the push-in connector labelled as Air + CO2 out on the rear panel of the DGT-CO2BX and to the preheating module or to the humidifying module or directly to the chamber, depending on the incubating system model. Figure 4: Zoom of the blue tube and silicon tube connection. Tip Look at the instructions written on the labels attached on the transparent plastic bags. Tip In order to avoid gas leak and correctly insert the tube, please, strongly push the tube into the Swift-Fit connector. 9

Tip Connect the 4mm I.D. tube to the humidifying Module or directly to the Incubating Chamber if the preheating module is not present. Tip Look at the colour of the tubes and of the adhesive stripes attached on these devices. Tip This tubing moves the Air and CO 2 gas mixture to the Specimen Chamber. 5.3.2 Connect Gas reservoirs with DGT-CO2BX Unit White rigid tubes (4): Connect the controlled (filtered and at P=1atm) flows of air and CO 2 (O 2 optional configuration) from Pressure Gauges provided by to the DGT-CO2BX. Insert the tubes into the Swift-Fit connectors (Air IN and CO 2 IN on the rear of the Unit, see Figure 3. Tip If you want to disconnect the tube, please, pull the tube while pushing the black ring on the Swift-Fit connector (Figure 5). Tip In order to avoid gas leak and correctly insert the tube, please, strongly push the tube into the Swift-Fit connector. 5.3.3 Fix the Filter holder to the rear panel of the DGT-CO2BX Unit Tip The DGT-CO2BX Unit is equipped with two additional Hydrophobic PTFE filters with degree of filtration 0.2 microns for the protection against bacteria and condensations. One of them has to be mounted on the rear panel of the control box. It filters the Air and CO 2 flows coming from their tanks before entering into the carbon dioxide sensing cell. It prevents sample contamination from bacteria and infra red CO 2 sensor from damages caused by dirty gas streams. It is connected with two 4 I.D. mm silicon tubes with the Filter In and Filter Out connectors on the Rear of the DGT-CO2BX. It is fixed by a support, called "Filter Holder" to the Control Unit. This "Filter Holder" is fixed to the rear of the control box with two screws. Refer to Figure 6 for the instructions about how to mount this Filter holder to the rear panel of the control box. Another Hydrophobic PTFE filter is present along the Blue marked tubing #1 tube composed by 1 meter of blue rigid tube of 6mm OD at each end, 50cm of silicon tube of 4mm I.D. with a filter, degree of filtration 0.2 micron. This filter avoid contamination coming from the Humidifying module to the sensing cell, done by a small back pressure, that may occur during the CO 2 or Air pressurized lines closing. Tip Replace the gas filters each 2 months of continuous use (Figure 1, number 4; Figure 6). Okolab has these filters at stock. Contact Okolab to purchase them. Do not remove the filters, absolutely. This may cause damages to the sensing cell and sample contaminations. Avoid narrowing of the tubes and formation of air bubbles inside them. Please, periodically check fluids are correctly flowing and tubes are properly connected. 10

Connect the Inlet of the the Filter to the Filter In connector on the rear panel of the control box. The inlet of the filter is labelled on the transparent plastic filter housing. 11

6 PC cables connections I. Connect the DGTCO2BX-PLUS to your PC using the RS232 Serial Cable supplied. II. Use the RS232/USB converter cable if your PC does not have RS232 serial communication port available. Please, install the driver on your PC using RS232/USB converter cable installation CD and re-start your PC. 7 Check your International options IMPORTANT For a proper work of the Okolab Read Temperature software, you MUST select the point as decimal separator in the International options of your PC, following this path: Start > Control Panel > Regional and Language Options > Regional Options > Customize Click Start. Select Control Panel. Select Regional and Language Options in the Control Panel. Click Customize in the Regional Options window. Select or write the point as decimal symbol in the text box. Select or write the space or comma as digit grouping symbol in the text box Click Apply than OK 2 times.. 12

Figure 7: Regional and Language Options Panel 8 Install the Software Insert the Installation CD-rom on your PC. Launch the setup.exe and follow the instructions on the screen. Once you have completed the installation, the program will appear on your PC Programs Menu. Re-start the Pc. 9 Change the Set Point The electronic controller has two different Set Points. Te Set Point 1 (SP1) represents the controlled CO 2 value and the Set Point 2 (SP2) represents the desired steady state value for Alarm functions. The Unit normally displays the current CO 2 concentration and the Set 13

Point 1. The SP1 is 5.00%, as default. SP2 has to be equal to SP1. If you wish to change manually the two Set Point values, please read the following instructions: Figure 8. Electronic controller display. IMPORTANT Do NOT change others parameters already adjusted to optimize the performances of the Unit. Press 1) Press, if necessary until SP1 prompt appears. Press 2) Display shows previous value of Setpoint 1. Press & 3) Press and to increase or decrease Setpoint 1 respectively. Holding & buttons down for approximately 3 seconds will speed up the rate at which the Setpoint value increments or decrements. Press & 4) Continue to use and to enter your 4-digit Setpoint 1 value. Press 5) Display shows STRD stored message momentarily and then advances to SP2 only, if a change was made, otherwise press a to advance to SP2 Setpoint 2 Menu. Do the same for SP2. The software supplied by Okolab allows to change the Set point during an experiment and to change the alarm configurations. It reads the Carbon Dioxide percentage in time and stores the data in the computer memory. Ask to Okolab for further informations. 14

10 Starting the experiment and Getting the best out of your incubation system 10.2 Organize the experiment Read carefully the manual of the Incubating System you are using to control the specimen temperature. If the temperature controller has been supplied by Okolab, ask to us how to get the best from the incubating system equipped with the DGT-CO2BX. IMPORTANT Overloading the Humidifying Module could induce condensation inside the tubes, on the windows surface of the incubating chamber and on the surfaces of plastic supports, not depended on the DGT-CO2BX. Avoid gases back flow. Check that all tubes are properly connected and prevent narrowing, throttling, kinking of tubing, and formation of air bubbles. Check the tubes are well fixed so they cannot slip off using the kit supplied. Switch on the Temperature Controller. 10.3 Air Flow Meter regulation IMPORTANT Do not use the flow meters as closing-opening valves. Once regulated the flow rate (at P=1bar, see Figure 3, use the flow meters carefully only for fine regulations. Open the Air gas valves slowly, regulate the pressure at 1 bar. Otherwise turn on the Air Pump with filter inside. Regulate the floating ball of the Air Flow Meter within the range 0.3 0.8 Litres/min. 'Target Gas Percentage indicates the stream gas percentage that the operator wants during the experiment. Air flow rate values and CO2 consumption, in Normal litre per minute [NL/min], are the values of air flow rate set on the graduated scale and the corresponding CO 2 consumption. 'Total indicates the measured total gas stream flow rate. Regulate the Floating ball Air Flow Meter at the desired value within the range 0.3-0.8 NL/min. Open the pressure gauge of the CO 2 reservoir and set it at 1.5-2 bar. Regulate the Pressure Gauge for CO 2 with regulator, 20 micron filter inside, supplied by Okolab at 0.9-1 bar. Be sure that no pressure drop happens. Use this pressure regulator as closing valve when you finish the experiment too. Turn on the DGT-CO2BX. System has been calibrated at the factory. It is maintenance free. Sensor is stable up to five years of work. We suggest to calibrate the Unit each two years of continuous use. Contact the Okolab Assistance to do the calibration. 11 Appendix 11.2 DGT-CO2BX parameters The PID closed loop controller reads the CO 2 level within the gas stream and gives feedback to a fine valve regulating CO 2 flow with an accuracy of ± 5% of CO 2 percentage. The PID parameters were stored to obtain the maximum performances from the system in terms of stability and fast recovery, after a gas flow changing due to quickly changed of pressures. The controller needs half an hour to reach the steady state after turning on the unit. Avoid modifying the controller factory parameters. Some parameters alterations could make the system bad-functioning or could induce the system failure.
