3 This manual details the steps necessary to install a DART source on all Bruker atmospheric pressure inlet mass spectrometers (except Bruker Esquire). This document uses a DART SVP 100 or 200 series source; however, the instructions should also provide general guidance for installation of a DART 100 or 100S. 4 Installation of the Bruker Vapur Interface For TOF instruments it is suggested to calibrate the instrument before switching to the DART ion source. Vapur Interface Installation 1. First, place the instrument in standby mode and remove all gas and electrical connections from the existing Bruker ESI (or other) source. 2. Remove the Bruker ion source and carefully place it to the side or in storage. Remove the Bruker end cap and atmospheric pressure inlet (API) inlet splash guard from the atmospheric pressure inlet area. The end cap and splash guard are pictured in Figure 1. a. CAUTION: These pieces will be extremely hot. Use gloves or wait for capillary and source temperatures to cool before removing. Figure 1: Bruker end cap and splash guard removed. B) The liquid drainage barbed fitting under the API inlet is sealed with the fitting provided by IonSense and the liquid drainage tube is slid over the fitting in order to seal the tube. 4

5 Figure 4: Vapur Interface in place on Bruker maxis 4G in the open position. Note: The latching bracket on the Bruker instrument will need to be adjusted for the DART Vapur Interface. To adjust the length of the latch, loosen the thumb nut and then rotate the entire latch to lengthen the latch. Tighten the thumb nut to lock the latch length. Inserting the Ceramic Ion Transfer Tube into the Vapur Interface Remove the Swagelok nut from the front of the Vapur Interface and insert the ceramic ion transfer tube into the interface. Secure the ceramic tube with one of the provided graphite Vespel ferrules making sure to set a 2 mm gap inside the interface. Note: To easily set the 2 mm gap, push the ceramic tube against the Bruker capillary, secure the Swagelok fitting with ferrule and then mark the tube with a pencil. Pull the tube out as shown in Figure 5 below so that a 2 mm gap is created inside the interface. Figure 5: Ceramic ion transfer tube set correctly in the Vapur Interface. 6 Attaching the DART to the Vapur Interface Once the Vapur Interface has been successfully installed, you are ready to attach the DART source. Refer to the DART SVP Hardware and Network Installation Manual (pages 6-8) for more specifics on interfacing the DART SVP source to the Vapur Interface. IonSense website: ( 6

7 Addressing Software Vacuum Reading Errors Because the mass spectrometer is not designed to accept the large flows of helium gas required to run the DART, you may receive errors informing you that the Bruker vacuum is too high. This is not indicative of system damage and will not damage your instrument. It simply means that the vacuum gauge, which is calibrated for nitrogen gas is giving a falsely high readback. The error message can be addressed by changing the upper vacuum limit for the fore vacuum gauge. To do this, perform the following procedure: Go to the Service tab in the Bruker software: Select the MPC 555 Board from the list on the left side. _ Then enter the command VACU:SRPS=10 in the "Command" line at the bottom of page. Note: 10 mbar is the recommended value for the fore vacuum upper limit value. The Bruker default factory setting is 5 mbar on the maxis. Click "Send" button on the page to submit the command. Other Useful Commands "VACU:HELP?" shows a list of all commands. Enter "VACU:SRPS?" to see what the current pressure threshold is set too. It should be set to 10. 7

8 Recommended Operating Parameters Ion Source: Turbo Spray Bruker source settings in the software: End Plate Offset: 0 V Capillary Voltage has no effect on signal intensity set to 0 V Dry Gas: 0 L/min o The signal intensity was approximately 10 x lower when the dry gas was on and set at 4 L/min than when the Dry Gas was set to 0 L/min The Dry Gas Temperature has almost no effect on the DART signal. 8
